Genotoxicity Testing: a Review
نویسنده
چکیده
Genotoxicity is a major limitation of various compounds used in clinical practices which range from gene mutations, chromosomal aberrations to other DNA effects. For the same reason, it must be assessed before bringing into the market. Moreover, various regulatory agencies have made it mandatory to submit the genotoxicity data while filing a New Drug Application (NDA). This in turn has given further importance to genotoxicity tests. This review deals with newer testing techniques used in genotoxicity in comparison with the established standard techniques.

A Novel Strategy to Predict Carcinogenicity of Antiparasitics Based on a Combination of DNA Lesions and Bacterial Mutagenicity Tests
Genotoxicity and carcinogenicity testing of pharmaceuticals prior to commercialization is requested by regulatory agencies. The bacterial mutagenicity test was considered having the highest accuracy of carcinogenic prediction. However, some evidences suggest that it always results in false-positive responses when the bacterial mutagenicity test is used to predict carcinogenicity. Along with maj...
متن کاملOecd Guideline for the Testing of Chemicals
1. The OECD Guidelines for the Testing of Chemicals are periodically reviewed in the light of scientific progress, changing regulatory needs and animal welfare considerations. The original Test Guideline 487 was adopted in 2010; it has been revised in the context of an overall review of the OECD Test Guidelines on genotoxicity. A document presented as an Introduction to the Test Guidelines on g...

Ever since the introduction of the Salmonella typhimurium mammalian microsome mutagenicity assay (the 'Ames test') over three decades ago, there has been a constant development of additional genotoxicity assays based upon the use of genetically engineered microorganisms. Such assays rely either on reversion principles similar to those of the Ames test, or on promoter-reporter fusions that gener...

With the need to understand the potential biological impact of the plethora of nanoparticles (NPs) being manufactured for a wide range of potential human applications, due to their inevitable human exposure, research activities in the field of NP toxicology has grown exponentially over the last decade. Whilst such increased research efforts have elucidated an increasingly significant knowledge ...
متن کاملGenotoxic and carcinogenic effects of gastrointestinal drugs.
This review provides a compendium of retrievable results of genotoxicity and carcinogenicity assays performed on marketed gastrointestinal drugs. Of the 71 drugs considered, 38 (53.5%) do not have retrievable data, whereas the other 33 (46.5%) have at least one genotoxicity or carcinogenicity test result. Of these 33 drugs, 15 tested positive in at least one genotoxicity assay and 13 in at leas...
